Please assist. The sliding door behind the driver's door does not open with remote button or the switch inside the car. Can only open manually now. Although it waas working remotely before.

I have had the same issue recently. First it was the passenger side. It started while we were at the beach. After we left the beach it started working fine on that side but then the driver's side started sticking. I believe that two things are associated with it. One, is that the salty, moist air has caused some stuff to get 'sticky'. The other is that we have driven in dusty/dirty conditions, heavy rains, and have had the car washed a number of times. All of these things taken together likely have affected the lubrication of the track and latch system. I'm going to take mine to the local Toyota Dealership and see if I can get everything greased and lubed. Hopefully that will work. There was also a recall notice a few years ago for issues with the sliding doors sticking or malfunctioning on certain models. Ours is a 2005 XLE Limited AWD. The recall was on our model but because we weren't actually experiencing the problem at that time, they didn't want me to bring it in. You might want to check your model by the dealership and see if it fits the recall. If it does, the dealership will take care of it, warranty or not.
