What could cause the tranny dipstick to read empty and tranny go out with the exception of reverse. Then two days later not being moved dipstick reads full and the gears work again?

The proper way to check the transmission fluid level is when the car is warmed up, on a level surface and in park. Did you check the level with the above conditions both times? If not, you will see a difference.
