When was the last time you had a tune-up? Could be a bad spark plug, faulty injector, or a problem wit one of your ignition coils. It is never a good idea to run a car while it is misfiring. It will cause damage to your exhaust system.

100% correct, that is why the check engine light is flashing. The only thing not mentioned was low compression on that cylinder. As soon as the misfire is fixed the MPG will go back up.
