Can i supercharge my 78 c10 with a 250 inline 6

Yes you can, but why? it's not that good of a motor to be handling any kind of boost, you will most likely scatter it all over the road. As with most any American in line 6 cylinder they were designed to be torque engines and had a much lower redline than a V8.

For hop up a parts on these inline engines, look up Clifford Research. You would probably be better off trying to turbocharge an engine like this, but would limit boost to about 6 p.s.i. unless you rebuild the engine with forged.pistons and other parts to beef it up. If you spend too much, you're better off just dropping in a small block V-8.
