Could be a solenoid opened or shorted, transmission control unit not sending the shift signal. Is your check engine light on?

Yes my check engine light is on

Have the codes checked at your local parts store, write then down then come on back here for further diagnosis, Buicks have a lot of solenoid problems, you can research them on the Internet. Costs have been running around a grand for the repair, some cheaper some higher.
