Dodge ram shaking rear end

2006 dodge ram 1500.
New rear u-joints, brakes are good,
checked fluid in rear differential and
transmission.  Truck shakes when
(sometimes) going uphill or under load
at 90km/h.  Does not happen if I'm in
gear 2 instead of drive/overdrive.  Is
my transmission on the way out?  It is
not consistent. Anyone got some bad
news for me?

I have a 2006, 1500. When putting it in gear or drive it makes a clunck noise. The same going from drive to reverse. When driving at speeds of 45-60mph a vibration not so much at higher speeds. I've noticed it has a roar of such inside the truck when driving and vibration tends to go away when I let off the accelerater. New tires. Shifts smooth. I'm trying to get this truck right as it is my first and my daily driver to take my grandsons to and from school. The moter has high miles but has been very well maintained and is smooth. Please someone has to have the answer to my vibration concerns???
