The arrival of 2006 IS 350 signified the beginning of the compact sedan's second generation. While all ISs had been categorized as sports sedan, the 350 was seen by many to be the first IS to live up to that name. The IS 350 had borrowed its suspension system from the high performance GS series, and its 3.5 liter engine was the largest ever dropped into the small sedan.

The new aluminum block engine replaced the inline six cylinder that was utilized by all prior ISs. This 3.5 liter 32-vavle DOHC power plant supplied 306 horsepower and 277 pound feet of torque, and the car weighed just 3500 pounds. The IS 350 rocketed from 0 to 60 mph in just 5.6 seconds and it turned the quarter mile in 14.5. Strong brakes halted the sedan from 60 mph in just 119 feet. The 350 was not quite as fuel efficient as the 250, but with a rating of 21/28, it was surprisingly close. This upper tier IS was available only as a six-speed automatic, rear-wheel drive sedan.

Owners and drivers found this new IS to be an incredibly impressive car: an ideal combination of power and handling. The new interior had a much finer fit and finish, but many owners complained about excessive rattles at high speeds. A bulk of other issues were related to optional equipment. Many found the Bluetooth and navigation systems difficult to operate, and learning the voice command system was also considered a considerable challenge.
